1). Fabricate a range of optimised substrates and demonstrate the presence or absence of aging effect upon performance and, in as much as possible, within a 12 month programme to look at the stability of substrates prepared earlier in the programme.(2). Prepare ranges of chemicals (oligo-nucleotides) with relevant anchor and linker to create a chemical array library and attach drops of specific probe molecules to local areas of a SERS substrate in a 2x2 matrix.(3). Explore ways to prevent the non-specific adsorption of analyte by covering non-probe surface Ag sites with inert molecules, e.g. mercaptopropane. {points (2) and (3) are linked}(4). Analyse the observed SERS spectra both before and after analyte presentation. Look for distance dependence effects by tests with different target species (e.g. ss and ds oligo-nucleotides with different chain lengths). Measure any variations on SERS arising from analyte presentation (e.g. pH and salt concentration).(5). Demonstration of spatial selectivity.
