Under this fellowship, I propose to continue to develop a variety of research programmes in the general area of synthetic organic chemistry with particular reference to heterocyclic systems possessing ring strain. I plan to develop new methods for organic synthesis using the rather unusual methyleneaziridine ring system. This work is expected to provide new ways for making complex polycyclic ring systems which incorporate a nitrogen atom. Such structural motifs are common to many alkaloids and other medicinally important organic compounds. I also plan to undertake studies directed towards the total synthesis of a naturally occurring antibiotic called ficellomycin. This compound contains a highly strained and very unusual 1 azabicyclo[3.1.0]hexane ring system. New chemical methods will be devised for the synthesis of ficellomycin. Through collaboration, we plan to screen our synthetic intermediates for antibiotic activity. In this fashion we hope to learn precisely how this antibiotic elicits its biological effects at the molecular level and stimulate new research on the design of novel antibiotics.
