It is proposed to use novel chemistry developed for the functionalisation of hydrogen-terminated silicon to synthesise short oligonucleotides at crystalline silicon surfaces. The ability of the resulting surface-tethered oligonucleotides to interact via hybridisation with complementary DNA strands will be explored. Finally, the ability of these surface-bound nucleic acids to act as substrates to enzyme systems which produce chemical transformations of DNA, such as nucleases and ligases, will be investigated. In this way chemical and biological syntheses are to be combined and exploited as a novel approach for the fabrication of semiconductor surfaces derivatized with biomolecules which may then be used to template further chemistry. This research would contribute to areas such as molecular electronics, biosensor design, and fundamental studies on DNA-mediated electron transfer.
