We have discovered 3 complementary modes of N-protection: 4,4',4''-trimethoxytrityl, 4-azidobenzyloxycarbonly (azido-Z) and methyl-cobaloxime. These offer wide-ranging improvements in protecting group methodology. The research aims to develop the methodology with amines, polyamines, amino acids and selected heterocycles. Thus, we will seek methods to discriminate between the amino functions of spermidin and apply these to the synthesis of biologically important spermidine derivatives, eg trypanothionine. We will apply the cobalooxime methodology to solve regiochemical problems in heterocyclic chemistry. We will also seek fundamental mechanistic understanding in order to optimise selectivity. WIth substituted trityl systems we will determine the extent to which the reactions are specific and general to optimise the utilisation of azido-Z fragmentation using stable isotope labelling in conjunction with NMR. We will explore alkylcobaloximes with lipophilic alkyl groups to ensure that they form complexes with chloropurines that are alkylated in solution and to enable mechanistic studies to be conducted.
