The research programme will consolidate and extend our synthetic work on bioactive marine macrolides. This will involve the development of new and practical methods for the construction of these complex target structures, many of which exhibit promising antitumour activity but are in extremely scarce supply form their natural source. Emphasis will be placed on achieving a high level of control over the stereochemistry and size of the macricycle. As an extension of our previously supported work on swinholide A, the bistheonellide A (misakinolide) project will explore the kinetic selectivity of ring formation as a function of seco acid structure and reaction conditions. The scytophycin C project requires the elaboration of an advanced intermediate already prepared in our laboratory. Selective ring formation is again anticipated, followed by the introduction of the sensitive N-vinylformamide functionality. The third ropic focuses on the development of an efficient synthesis of the potent antitumour macrolide, aplyronine A. This will require further development of our dipropionate chemistry, regioselective macrolactonisation techniques, and a new proceudre for the introduction of the N-vinylformamide group.
