Summary on Grant Application Form
1. The structure and dynamics of serum transferrins (80 kDa), its recombinant lobes, and mutants will be investigated in solution using NMR spectroscopy. 2. Comparison between species (e.g. human, pig, rabbit) and use of 15N and 13C labelled proteins will aid the assignment of resonances. Resonances which can be used as specific probes of the N- and C- lobes will be identified. 3. Processes relevant to receptor- mediated metal ion, proton, and anion uptake and release, including interdomian hinge-bending, will be investigated. Metal ions will include those of physiological, toxicological and andiagnostic importance, including Fe3+ , Al3+, In3+, and Co3+. Both synergistic and non-synergistic anions will be investigated. Binding constants for anions will be determined. 4. Evidence for communication between anion and metal ion binding sites, and the glycan chains will be sought.
