The ring expansion[3.3] sigmatropic rearrangement of vinyl substituted ketene acetals and animals derived from 1, n-diols and aminoacohols will be used in a tandem metathesis/Claisen strategy directed toward medium ring lactones and lactams. Medium ring stereocontrol will be employed to functionalise these products stereo- and regioselectively. The cis-double bond produced in the claisen rearrangement will be functionalised stereoselectivity under allylic stereocontrol to give novel building blocks for the synthesis of polyketide and alkaloid natural products. A two directional ring fusion strategy will be developed to assemble fused polyether structures. This will be applied to the core nine-membered ring-containing fragment of the polyether toxin ciguatoxin.
