The aims of the proposed work are to advance our studies in the area of chiral lithium amide base chemistry. We seek to improve the fundamental understanding of these reactions, which in turn will allow us to develop more selective and powerful bases, and to broaden the applications of the chemistry to new and novel transformations. The proposed work involves an NMR study of chiral bases in order to clarify aggregation phenomena and salt effects, and to predict the likelihood of non-linear effects in chiral lithium amide mediated reactions. A new family of 1,2-diamine bases will be synthesised and tested, preliminary results have shown that a single member of this group of bases shows outstanding promise. The scope and limitations of two unprecedented transformations, involving episulfoxides and phosphine oxides, will be examined, with the aim of developing these reactions into mature parts of the chiral base repertoire. A new and novel type of enantioselective enolisation will be examined as a possible entry to the zaragozic acids.
