The zaragozic acids (squalestatins) have attracted great attention as potential cholesterol-lowering drugs, and also have anti-cancer potential. This project will build on previous model studies in our group to complete the first total synthesis of one member of the family, zaragozic acid D. Having demonstrated the feasibility of our approach with this first generation synthesis, attempts will be made to develop shorter synthesis with greater control over stereochemistry. This will require investigation of several new pieces of methodology. First, a stereodefined trienol ether will be synthesised and subjected to an unprecedented, triple asymmetric dihydroxylation procedure. This will allow incorporation of all the stereocentres of the core of the natural product in a single step. A final approach to be investigated will develop a new way to introduce the tricarboxylic acid portion of the natural products. Of more general importance, it will also examine a new cross-coupling strategy that is of potential wider application to the stereodefined synthesis of alkenes.
