We have recently described a new mechanism for the Mo (0) -catalysed allylic substitution and found indications that Mo(II) may actually be superior to Mo(0). The new Mo (II) complexes have been shown to catalyse allylic substitution (at 0 degrees) with much broader range of nucleophiles (e.g., sillyl enol ethers, electron-rich aromatics, and selected aromatic heterocycles) than Pd(0) and to have the potential to catalyse other reactions, such as Diels-Alder addition, ene reaction, etc. In short, these catalysts have the promise of being complimentary to the more expensive Pd and superior to other metal catalysts. We are going to further develop the catalysts by the rational variation of spectator ligands with the view of designing enantioselective catalytic processes. If successful, the results will be directly applied by SmithKline Beecham to solve specific problems in the synthesis of their new drugs that are now in the development stage, and to improve the synthesis of another one, currently being manufactured.
