The application of an unconvetional method of carbon-carbon bond formation using a light-initiated radical process will be investigated. This approach benefits from the cheap, technically clean controllable method of radical generation and has enormous potential for large scale synthesis in the chemical industry. At present this important type of reaction is rarely (if ever) considered by the industrial chemist for carbon-carbon bond formation and there is an urgent need for the development and utilisation of radical intermediates in bulk chemical synthesis. The project will address the feasibility of this method and help to change current thinking towards the application of radicals in the preparation of a variety of industrially important amino acids. Investigations will concentrate on applying this method to alkylate glycine to afford a novel approach to a variety of racemic and chiral amino acids. These studies will explore the generality and efficiency of this method on a multigram scale. Particular attention will be drawn to the application of this methodolgy to the synthesis of three biologically active amino acids (levodopa, melphalan and levothyroxin) of considerable industrial importance.
