This proposal aims to provide a unique, highly efficient and rapid synthesis of the manzamine alkaloids. The chemistry involves, as its key step, a powerful and stereocontroled intermolecular azomethine ylide cycloaddition. This process sets up two new rings and three new chiral centres in a single step, to provide the ABCE rings of manzamines A, E and F. Ring D will be constructed from this tetracycle and the total synthesis completed in what should be the shortest and most efficient route to date. The manzamines are an important class of marine alkaloids with significant biological activity as antileukemic and antibacterial agents.
