We propose to combine donor-acceptor templating (using neutral robust building blocks) with thermodynamically controlled ring synthesis to give chemically versatile poly-catenanes. Use of removable blocking groups and a ring fixing reaction will give access to well defined poly-catenanes with new architectures. More importantly, this specific catenane proposal is a vehicle for exploring a general strategy for covalent synthesis of nanoscale objects. We propose to exploit a hierarchy of three types of covalent bond formation for the programmed generation of large rings: kinetically controlled, irreversible reactions; Thermodynamically-controlled, reversible reactions (transesterification and imine-formation); and ring fixing reactions (imine reduction).
