Existing methods of treatment (calcium hydroxide) for treatment of gross dental caries involving pulpal exposure show variable success. The present proposal involves development of a novel approach to wound healing in these tissues developmental events are mimicked by presentation of Transforming Growth Factor- b1 in an alginate hydrogel to simulate the dental basement membrane, which controls dental cytodifferentiation during tooth development. The design requirements for this cellular engineering approach wound healing in the dental tissues will be addressed and the system evaluated in a human dental tissue organ culture to characterize cellular events during healing.
