Multivalent high affinity carbohydrate libraries will be prepared and evaluated for biological activity column chromatography. A range of disaccharides will be prepared using a recently developed latent-active glycosylation strategy. The disaccharides will be coupled to a spacer arm which enables attachment to an amino group containing arm of a polyamidoamine (PAMAM) dendrimer. A range of PAMAM supported disaccharides will be prepared which will mixed, split and coupled with vinyl glycosyl donors. The libraries of saccharides will be subjected to an affinity column and the structure of a retained compound elucidated by a combination of methods including enzymatic degradation, methylation/GH analysis and mass spectroscopy. Dendrimer supported oligosaccharides will be used to find new ligands for pathogenic entero-bacteria. In a later stage of the research, common allyl glycosyl building blocks will be prepared that can be used for the synthesis of branded saccharides. A monosaccharide will be attached to the dendrimer and two combinatorial glycosylations will be performed.
