Research will be undertaken in the general area of synthetic carbohydrodrate chemistry. New approaches to the synthesis of C-glycosides will be investigated. C-Glycosides are synthetic analogues of naturally occurring O-Glycosides in which the anomeric linkage and are thus represent possible drug targets as O-glycoside mimics with improved therapeutic profiles. The two approaches adopted should allow the stereocontrolled synthesis of a wide range of C-glycosides in high yield. Both approaches involve the use of precedented sigmatropic rearrangements. In particular it is hoped that these approaches be extended to allow the synthesis of both C-disaccharides and functionalised spiroketals. C-Disaccharides are analogues of natural O-linked disaccharides in which the interglycosidic oxygen has been replaced by a methylene group. The biological activity and therapeutic potential of such materials has yet to be investigated due to their limited availability.Functionalised spiroketals are structural units of many biologically active natural products.
