The azinomycins are a unique class of natural products that possess potent anti-tumor activity. Under this programme, we propose to draw on our existing expertise in this research field to complete the total synthesis of these complex and highly sensitive chemical targets. As no research group as yet accomplished this goal, the successful completion of the synthesis of azinomycin A or azinomycin B would represent a significant landmark in this internationally competitive field.In the proposal, we detail two novel approaches to the 1-azabicyclo[3.1.0]hexan ring system of the azinomycins which we believe will offer highly expedient routes to the azinomycins. Our general strategy relies on recognising that either the alpha- or beta-nitrogen atoms of the dehydroamino acid can be introduced by condensation of an amine (or amide) with the corresponding alpha-keto acid or beta-keto ester followed by subsequent imine-enamine tautomerisation. Further elaboration of the intermediates produced using this new chemistry should provide short routes to both azinomycin A and B.
