Characterisation of chemical and biological microstructure requires new optical imaging techniques capable of dynamic, non-invasive and quantitative analysis, in particular, in situ measurements under reaction conditions. The main scope of this project is to develop a novel analytical approach, interferimetric scanning microscopy (ISM), which will combine and expand the advantage of digital interferometry imaging modes, will allow for surface and volume principal configurations of the ISM, epi- and trans-illumination imaging modes, will allow for surface and volume phase measurements with nanometer vertical resolution at the micro-second timescale. Exploratory studies will be carried out for a protein system (insulin) in order to investigate the elemental molecular structures, surface growth kinetics and formation of crystal defects. In addition, flow-scanning mode of the ISM will be tested using particle formation process in supercritical carbon dioxide. This project will continue our long-term programme aimed to develop in situ optical analysis for chemical pharmaceutical science and will be supported from industry.
