This proposal is for a detailed physical organic investigation into the role of metal ions in catalysing phosphate monoester and diester hydrolysis. Several lines of study are proposed, aimed at measuring quantitatively the effect of thio substitution at phosphorus in this context, the effect of combining further catalytic modes of action and in creating a link between mechanistically useful ions such as Co(III) and biologically interesting ions such as Zn(II). This will be achieved through kinetic analysis of defined complexes, where the contact between phosphates and metal ion are known, and where the mechanism of action can be shown. Based on the initial results, we will be able to construct more complexe models, making efficient use of any synthetic effect. The results will be applicable to understanding important biological catalysts which utilise metal ions for catalysis and to designing better artificial catalysts.
