We have very recently discovered that model homoproparygylic glycinates, in which the amino group is masked as the corresponding sulfonyl derivative, undergo smooth overall 5-endo-dig iodocyclisations to give 4-iodo-2,3-dihydropyrroles in excellent yields. These are stable compounds which offer a wide range of possibilities for further elaboration; additionally, these should be available in chiral, non-racemic forms and should also be readily converted into the corresponding iodopyrroles. The first objective is to define the scope and limitations of the initial method by examining more highly functionalised substrates, alternative electrophiles and stereochemical integrity. Subsequent chemistry of the initial iodo- dihydropyrroles will be focussed mainly on their suitability as partners in a wide range of transition metal-catalysed coupling reactions. The resulting products will be useful in further chemistry, including Diels-Alder cyclisations, electrocyclizations and ene reactions. Annulation chemistry of the dihydropyrroles will also be examined with a view to setting the stage for a total synthesis of the unique macrocyclic antibiotic Roseophilin, which will form the theme of the second part of the project.
