We have recently disclosed a new method for the synthesis of bicyclic beta-lactams. This is based on novel azomethine ylid reactivity and the direct and flexible nature of the chemistry has important implications for the pharmaceutical industry and medicinal chemistry discovery ; two patents have already been filed. This proposal takes these initial observations and aims to develop new directions that will firmly establish this method by extending the scope of both the transformations available and the range of structures that can be prepared. Specific issues that are dealt with include the construction of a series of new and important oxazolidiones (as precursors of the azomethine ylid reactivity), less stabilised (and, as a consequence, more reactive) 1, 3-dipoles, and novel oxazolidionones that are designed to provide a direct and flexible entry to carbapenems - our current methods have not yet solved this important issue. We also want to apply the mechanistic information that we have accrued towards new and speculative transformations that, if successful, (and we have encouraging initial results), will further enhance the power of the underlying synthetic strategy.
