The proposed research covers a number of topics in biological and synthetic chemistry, most of which are concerned with the development of biologically active compounds. Thus the chemotherapy of cancer is being actively investigated with the development of new agents based on indolequinones, the azinomycins and manzamines; the key activation of indolequinones with reductases is also being studied. Diaziridine mimics of natural sugars will also be studied as will new routes to bioactive pyrrolidine alkaloids. A major study on thiopeptide antibiotics (synthesis and mechanism of action) will be initiated. Studies on the cloning, protein overexpression, biochemical characterisation and structural determination of a number of key enzymes involved in biotransformation reactions and of medical importance will continue within the protein structure group. In collaboration with this group it is proposed to investigate model complexes of the vanadium haloperoxidases, and supramolecular complexes of carboxylates, amino acides, and catecholamines. Novel liquid crystalline systems will also be investigated.
