There is a demand for a portable device for the measurement of pollutants in environmental waters with increasing Government legislation. While the surface selective technique of surface plasmon reonance (SPR) has proven to be an extremely useful technique in the biomedical field it has not been utilised in environmental monitoring as current commercial systems are large laboratory based instruments. While a hand-held SPR instrument is now available it can only measure non-specific interactions or changes in refractive index. The aim of this research proposal is to develop novel surface chemistries in order to formulate monolayers of natural receptors which specifically interact with target pollutant analytes. The molecular recognition molecules are three proteins which will be formulated as monolayers on gold coated waveguides via the process of self-assembly. Changes which occur at the monolayer suface upon interaction of the target analyte will be probed using SPR. In the first instance the SPR instrument will be a lab based system although the key goal of this research will be to develop, in collaboration with other partners in the MuttE consortium, a portable SPR instrument for in situ measurement of nitrate, nitrite, phenol and chlorinated phenols in raw and potable waters.
