A novel detection method is proposed for any analyte that can be selectively immobilised or absorbed from solution onto a smooth substrate. Analyte binding sites are arranged in a spatially periodic array, (preformed either by holographic or by soft lithography using an elastomeric stamp), so that when the analyte is bound the refractive index is locally (and thus periodically) modified to forma diffraction grating. The coherent scattering gives a signal proportional to the square of the analyte concentration, and is intrinsically far more sensitive than schemes that employ incoherent detection. The method uses inexpensive optical hardware common to all analytes and a cheap disposable sensing element to provide analyte selectivity. Target analytes include proteins, nucleotide sequences and trace contaminants such as pesticides.
