It is proposed to develop the chemistry of chiral oxime ethers derived from (R) - and (S0 -0- (1-phenylbutyl) hydroxylamine [ROPHy and SOPHy] in asymmetric synthesis. Thus it is proposed to develop an improved asymmetric synthesis of alpha-aminoacids, especially non-proteinogenic and alpha-alkylated aminoacids. We will also investigate the metallation chemistry of chiral oxime ethers, in particular alpha-lithiation and beta-palladation, as a method for the asymmetric alkylation of aldehydes and ketones and functionalisation of inert alkyl groups respectively. The use of ROPHy/SOPHy oximes in combination with the ring closing metathesis reaction (RCM) will be developed as a powerful route to chiral cyclic amines. Finally it is proposed to develop a solid phase variant of the reaction for use in combinatorial chemistry.
