In order to develop novel radio-pharmaceuticals we wish to synthesise new transition metal complexes which have reasonable stability in vivo. In our preliminary work with PS2 and its analogue/homologues we have demonstrated unique properties most of which arise from enhanced complex stability associated with the favoured endodentate macrocycle conformation. Use of this soft ligand in pharmaceutical applications with soft metal centres will reduce competition with the mainly hard metal ions and their associated ligands present in serum. The project will start by rapidly preparing and characterising a range of new PS2 complexes. Attention will focus on those elements of relevance to diagnostic and therapeutic nuclear medicine (Tc, Re, Cu, Ag, Hg, Au). Stability of the new complexes in serum will be assessed. The project will also develop new carbon-functionalised chemistry of PS2 and its benzo analogue in order to control the properties of the new complexes for application as metal-essential radio-pharmaceuticals and also to synthesise bifunctional derivatives of PS2 and use them to synthesise conjugates of biomolecules (the metal-tagging approach).
