To date we have established a flexible route to the carbon skeleton of the anti-tumour, cyclic peroxide containing, sponge norsesterterpene, mycaperoxide B. However, attempts to introduce the peroxide unit, based on a biogenetic route proposed by other workers have failed, calling into question the original proposal. In light of a chemically more plausible proposal we have modified our approach and intend to study its application to completion of the total synthesis. This will require particular attention to stereo- and regio-control. Successful application of this second approach will lend support to the second biogenetic proposal and this methodology can then be applied to the synthesis of closely related members of the mycaperoxide family to confirm somewhat speculative structures based upon sometimes ambiguous analytical data.
