We have discovered a user-friendly method for preparing aziridines which is mediated by catalytic amounts of sulfides and metal salts and stoichiometric amounts of imines and diazoprecursors (reactive diazocompounds are intermediates but are never handled). However this has only been conducted with simple sulfides and N-Ts imines and at present the diastereoselectivity is low. There is now a need to improve the diastereoselectivity of the process, to exploit the in situ generation of diazocompounds for catalytic asymmetric aziridination, to explore alternative diazocompounds and to apply the methodology to the synthesis of biologically important targets. The diastereoselectivity will be optimised by variation of the groups on the imine nitrogen. Those that show high diastereoselectivity will be tested in asymmetric aziridination using sulfides available from our catalytic cycle for epoxidation. Successful diazo precursors (e.g. aliphatic, alkenyl, alkynyl diazocompounds) used in epoxidation will be tested in aziridination together with other more stable diazocompounds (Me3SiCHN2, diazophosphonates, diazoesters and diazoacetamides). Success in this project will provide a general method for asymmetric aziridination and this will be showcased by the synthesis of biologically important targets.
