1, 2-Metallate rearrangements involving higher order cuprates of metallated enol ethers offer a powerful method for the stereoselective synthesis of trisubstituted alkenes. A preliminary investigation has revealed new conditions which overcome serious limitations to the scope of the reaction. The new conditions will be optimised and deployed in the synthesis of dienes and trienes involving carbohydrate glycals as reactin partners. With the aid of mechaistic investigations, a catalytic variant of the reaction will be explored in the search for maximum economy, efficiency and scale. Further enhancements in efficiency include the use of tandem (Cu(I)-catalysed processes (eg carbocupration of alkynes) terminated by a 1,2-metallate rearrangement as a means of stereoselective construction of polyenes involving the formation of up to 4 C-C bonds in one pot. The synthesis of some simple biologically active natural products will illustrate the advantages of the methods.
