Under this Foresight LINK programme we will develop fabrication methods that generate porous scaffolds with two distinct types of porosity: anisotropic and random. Each scaffold will have an anisotropic component (eg an aligned channel for fibre) that will run across the thickness of the scaffold. This component will align cells early in the tissue regeneration process and provide routes for nutrient supply. In addition, the random pore network will encourage nutrient supply throughout the scaffolds.Scaffolds will be produced from both biodegradable polymers and ceramics using a number of fabrication methods. The scaffolds will undergo a series of physical, chemical, mechanical and surface characterisations. Suitable scaffolds (based on pore distributions and anisotropic structures, surface properties and mechanical responses) will be carried forward for screening using cell culture models. On the basis of the ability of the scaffold to encourage rapid cell migration, extended cell viabilities across thick scaffolds and integrin-mediated spreading we will select scaffolds for bioreactor studies. These studies will aim to demonstrate that substantially improved cartilage and bone tissues are created by the presence of the unique pore distributions in the novel scaffolds.EngProg\
