This proposal targets, for the first time, dendritic macromolecules in which an anionic guest is bound at a single, well-defined binding site encapsulated within the branched superstructure. This approach is intended to model anion binding enzymes, which possess deeply buried guanidinium subunits.The dendritic shell will play an active role in the recognition event. For example, by tuning the solubility of the receptor, and hence the host-guest complex as a whole, the branched shell will enable the extraction and transport of biologically important hydrophilic anions. The dendritic branching should also shield the recognition site from bulk solvent, thus diminishing the effect of solvent on the thermodynamics of simple anion recognition - leading to a truly unique microenvironment in which molecular recognition can occur.Furthermore, we intend to investigate the ability of these new functional dendrimers to act both as anion sensors and catalysts for phosphate hydrolysis. In all cases, the information generated about the unique effects of the branched shell on molecular function will be used to assist in the design of more advanced dendritic anion receptors, and eventually in the development of novel functional devices.
