Summary on Grant Application Form
A combination of synthetic chemistry, electrophysiology and X-ray crystallography will be used to investigate the potential of oxacalix[3]arene derivatives to induce sodium permeable transmembrane channels. Derivatives of oxacalix[3]arenes will be prepared and characterised by standard analytical methods including X-ray crystallography when possible. The abilities of these compounds to facilitate transmembrane cation transport across both artificial bilayers and whole cells will be analysed by patch clamping and single channel recording techniques. Information concerning the selectivities of these compounds for sodium (in the presence of other bioavailable ions) will be gathered and the mechanism elucidated. Results form this study will give greater insight into the mechanism of cellular cation channel formation and the factors which influence selectivity and transport.
