Summary on Grant Application Form
Phase 1 of the programme will involve developing and improving the methodology developed by us for the asymmetric synthesis of alkaloids of the ajmaline family, starting from L-tryptophan. It is intended that a number of paths will be explored in the earlier stages, to provide access to a key advanced intermediate.Phase 2 of the programme will involve the exploration of a completely new route to the ajmaline family of alkaloids using carbene insertion/cyclopropyl chemistry. We aim to prepare an advanced intermediate with 5 of the key rings of ajmaline in place.In Phase 3, we expect to complete the total asymmetric synthesis of at least one member of the ajmaline family.....hopefully ajmaline itself. However, if phase 2 does not appear to be generating the framework we require, we expect to include raumacline and Alkaloid G as additional targets using the methodology developed in Phase 1.
