There is a strong perceived need for the development of novel bone cement materials for total hip replacement operations, particularly in terms of developing strategies for effective antibiotic loading and release. In this investigation we intend to systhesise a range of quinolone conjugated polymethylmethacrylate (PMMA) derivative and to examine the in vitro properties of these materials in terms of drug release behaviour, physical properties and the ability to prevent biofilm growth. More specifically we plan to synthesese nalidixic acid and ciprofloxacin derivatives of PMMA using a hydrolysable ester spacer group and an intramolecular nucleoplile neighbouring moeity in order to facilitate control of the drug release rate. Dissolution testing will be performed along with physical assessment using dynamic mechanical thermal analysis, tensile testing and modulated temperature DSC. Biofilm formation on disks of the candidate materials will be examined using the Robbins device.
