The proposed research deals with the development of bifunctional asymmetric catalysts for a number of key organic transformations. This emerging area of research promotes asymmetric synthesis by the activation of both components in a reaction and is inspired by the multifunctional catalysis frequently observed in enzyme promoted transformations. We shall design novel, metal free, bifunctional catalysts for key organic transformations based upon the linking of two catalytically active nitrogen heterocycles. Addition of a second linkage will produce macrocylcic bifunctional catalysts in which the reaction will take place in a pocket between the two nitrogen centres. Asymmetry will be induced by the presence of chiral bridging groups between the catalytic portions. This separation of catalytic centres and asymmetry inducing portions of the molecules makes the catalyst modular in design, simplifying the synthesis of a range of catalyst structures and facilitating the optimisation procedure. We shall design and investigate the use of bifunctional catalysts in several key areas of organic synthesis: halocyclisations, acylation and sulfonylation, Robinson annelations and Baylis Hillman reactions. -2-
