The biocompatibility of the currently available coronary stents is compromised by thrombus formation and excessive intimal proliferation leading to in-stent stenosis. In addition the highly corrosive in vivo environment results in gradual release of metal ions into the blood stream. A significant improvement of stent biocompatibility is needed to reduce the rate of intrastent restenosis and target vessel revascularisation. Such an improvement can be achieved either by (1) surface modification of currently existing stents; (2) novel stent design, or by (3) using new materials for stent design. This multi-centered interdisciplinary project involving surface engineers, chemists, cell biologists, material scientists, clinicians and industrial partners aims to address these problems through the development of novel stent coatings with improved biocompatibility. The project will involve an iterative process of stent coatjng and in vitro bioevalaution to determine the relationship between surface interfacial properties and material compatibility in this application. Optimised systems will be evaluated in a limited animal study to provide preliminary data to support subsequent product commercialisation through the industrial partners.
