The aim of this project is to complete the first total synthesis of the pestalotiopsin natural products using innovative chemical methods. Pestalotiopsin A was isolated only recently and has been found to display promising immunosuppressive activity and cytotoxicity in preliminary studies. Furthermore, the complex structure of pestalotiopsin A is unique among natural products and its construction represents a significant synthetic challenge. To date, no synthetic studies on the Pestalotiopisns have been reported in the literature. We wish to apply chemistry recently developed within our group in conjunction with new methods which we will explore during the course of the project, to build the natural products. Our preliminary synthetic studies show our approach is feasible, and that our synthetic goals are achievable in the timescale proposed. The new chemistry and understanding which will arise from our work will prove useful to synthetic organic chemists worldwide.Finally, as a result of our studies, both the natural products themselves, and analogues of them, will be available for biological evaluation.
