Enzyme-catalysed radical reactions are important processes in biology, whereby the enzyme must exert control in order to manipulate the reactivity and selectivity of reaction, often in the presence of highly reactive intermediates. We propose that proximal aromatic residues are involved in the fine control of reactivity and selectivity of radicals in enzyme systems. A quantification of this role will contribute to a predictive understanding of factors that control radical reactivity in enzymes and provide insights into how reactions as yet only observed in enzyme systems can be achieved in the laboratory.This proposal aims to address the issue of radical stabilisation by aromatic moieties through a multi-faceted approach. Paracyclophane and related model compounds will be synthesised and radicals generated with restricted geometries. The kinetic and spectroscopic properties of these radicals will be measured and correlated. These correlations will be contrasted with information on the stability of the radicals obtained through high level computational studies. This will then be consolidated in order to provide a complete description of the definitive factors ( with respect to aromatic residues ) that are employed by enzymes to control and direct free radical reactions and allow elucidation of mechanistic aspects that are currently poorly understood.
