The proposed research will implement a cw evanescent wave cavity ring-down absorption technique to study the structure and dynamics of interface molecular processes. The cw experiment will produce sensitivities allowing sub-monlayer dynamics to be observed by direct absorption. The binding characteristics of fused silica surface will be studied by measuring the binding of positive, negative and neutral molecules to the surface. Measurements as a function of pH will allow the number of surface SiOH groups to be determined. Experiments to control the surface bonding interactions will be investivated by passivating the surface with functionalised silylanes. The bonding characteristics of these new surfaces will be explored. Linear dichroism experiments will be performed to measure the angle or orientation of the molecule with respect to the surface. Measurements will be made on random, ordered and self-assembled layers. A change in the distribution of the angle will be observed in passing from a random phase to an ordered phase. Surface electromagnetic enhancement and surface plasmon resonance effects will be investigated on gold surfaces as possible signal transduction processes. The range of the enhancement will be determined in a thiol layer by varying the length of an alkyl chain between the surface and the chromphore. Estimates of sensitivity will be performed.
