Simple 1 chloroalkylmagnesium reagents possess exceptional configurational stability and have been recently synthesised by Hoffmann in enantioenriched form by sufoxide ligand exchange. We aim to investigate the insertion reactions of such carbenoids with boronic esters arid to develop an iterative asymmetric synthetic technique based on the anticipated reagent controlled homlogation processes. The proposed methodology is totally programmable and offers a degreeof efficiency riot possible with existing substrate controlled asymmetric boronic ester homologation technology. The research programme Will place an emphasis on the development of carbenoid reagents (1 chloroethylmagnesium bromide arid chloro(dimethylphenylsily)methylmagnesium bromide) suitable for the __asymmetric elaboration of polypropionate and polyol chains; structural features inherent to many important biologically active natural products arid particularly evident in the polyketides. The reagent controlled asymmetric homologation reactions of polymer supported boronic esters will be investigated at a later stage of the project. Finally, the methodology developed during earlier phases of the project will be exploited to synthesise a significant fragment of the complex polyketide discodermolide.
