This Proposalinvolves a completely new approach to dissolving biomolecules in supercritical CO2 (scCO2) using hydrophobic ion pairing (HIP) by CO2-philic surphactants. Preliminary results are very encouraging. Our objectives are: (i) to demonstrate in situ catalytic activity of enzymes while dissolved in scCO2 (ii) to identify a range of less expensive HIP surfactants for use in CO2 and other supercritical fluids (iii) to establish whether the HIPapproach can be extended to DNA. We believe that the project is adventurous; it combines wide academic interest with significant industrial benefits, ranging from fine chemicals manufacture to environmentally more benign dry-cleaning. Our application is backed by unrelated industrial funding.
