A single ion channel is usually modelled by a finite state space continuous time Markov chain. The state space is partitioned into conductance clas and it is possible to observe only which conductance class the channel is in. Short sojourns of the channel in any conductance class may be undetected. This phenomenon, which is known as time interval omission, hampers inferences concerning ion channel gating mechanisms from observed single channel recordings.In many single channel experiments, it has been observed that single channel recordings show periods of repetitive open channel activity, known a bursts, which are noticeably separated from each other by periods of inactivity. The aim of the proposed research is (i) to develop pseudo-likelihoo based methods for estimating single channel gating parameters, which are based on properties of bursts (such as total charge transfer) and are rot to time interval omission; and (ii) to apply these methods to MscL (mechanosensitive channel of large conductance) recordings.
