This proposal aims to couple procedures for the generation of samples which are enantiomerically enriched, but of extremely low optical purity using intrinsically chiral circularly polarised light (CPL) with those for enriching the enantiomeric purity of a sample by crystallisation methods. To this end we will synthesise a range of crystalline, photo-racemisable compounds similar to those which have previously been shown to be good candidates for deracemisation with CPL. We will investigate the deracemisation by crystallisation (asymmetric transformation, AT) of these compounds under conventional conditions (i.e. seeding their rapidly racemising, supersaturated solutions with single enantiomer crystals) and identify which compounds undergo AT, then attempt the AT using CPL to tip the equilibria of the photoracemisations. We will also synthesise and investigate the AT crystallisation of a range of derivatives of commercially important compounds, both with the use of CPL and by conventional AT (i.e. seeding or rendering enantiomers diastereomeric with chiral additives). Crystallographic studies of all the racemates will be undertaken to identify those compounds which are likely to be suitable candidates for AT deracemisation and a range of designer chiral acids will be synthesised to facilitate AT.
