The proposed research will involve fundamental studies on the structural properties of each of the polysaccharide systems IN described in the Objectives above, the molecular interactions involving them which underpin their current - and potential - therapeutic roles, and, where appropriate, how these therapeutic roles can be enhanced by chemical modification.The fundamental studies will involve purification followed by a combination of rigorous chemical and physical characterisations and correlation with biological activity. The UK (Nottingham) laboratory will direct the project and be involved specifically with (i) detailed physico-chemical characterisation (molar mass averages and distribution; sedimentation coefficient and distribution; intrinsic viscosity and distribution, radius of gyration and distribution, Mark-Howuink-Kuhn-Sakurada conformation coefficients, conformation assignment, flexibility and branching information) (ii) molecular interaction studies (nature of interaction - if any - stoichiometry, reversibility, strength/molar association constant, Ka etc.). These studies dovetail in with the isolation, purification and modification studies, and biological activity studies to be undertaken by the other European partners,: the collective results will help with the design and production of better and more efficient adjuvant and therapeutic systems.
