Established routes to complex polyether natural products involve the painstaking, iterative synthesis of the individual ring systems. Many of these natural products contain embedded centrosymmetric fragments. Despite this, reported syntheses have not exploited this hidden symmetry. We will exploit the desymemtrisation of a centrosymemtric molecule for the first time in a total synthesis: an enatioselective C-C bond-forming reaction will be used as a key step in a convergent, efficient synthesis of Hemibrevetoxin B.
