This proposal outlines research in organic chemistry, involving chiral organometallic species. We have developed a highly efficient, asymmetric synthesis of 2-substituted pyrrolidines by dynamic resolution of 2-lithiopyrrolidines. The aim of this research is to build on and exploit this remarkable result. The first priority is to convert our organolithium species to other organometallic species and to determine the stereoselectivity in reactions with these derivatives and the scope of electrophiles that can be used. The generality of the chemistry will then be probed, by extending the chemistry to important substrates, in particular for the preparation of enantiomerically enriched heterocyclic compounds such as 2-substituted piperidines, indolines, piperazines and tetrahydropyrans. Extension to acyclic compounds will provide chiral amines and alcohols. During this work, we plan to determine the rate of racemization of these chiral organometallic species and hence gain insight into the role of the substituents, solvent, temperature, additives, etc. on the chemical and configurational stability. In addition, we propose to explore a variety of methods for the formation of chiral organometallic species and the factors that influence the feasibility of dynamic resolution.
