Chiral nitrogen heterocycles are widespread in biologically active molecules, both natural and man-made. The intramolecular aza-Wittig reaction between an iminophosphorane and a carbonyl compound can be used to prepare cyclic imines of varying ring-sizes. We have developed an asyn variant of this process, whereby dicarbonyl compounds containing a symmetry element are desymmetrised during the ring-closure by way of a ch phosphorus reagent. This approach allows rapid access to polyfunctional cyclic imines and derivatives and has great potential for the concise as of relatively complex cyclic amines. The proposed work aims to build on the initial results by (a) optimising the reagents to achieve maximum asymmetric induction; (b) developing catalytic variants by in situ reagent recycling or azide-free iminophosphorane synthesis; and (c) demonstrati broad scope of possible structures accessible through this chemistry, and applying these results in the synthesis of representative small target molecules.
