We have recently developed novel one-pot procedures in York which allow rapid access to a range of C-glycosides. In preliminary studies we have utilised this chemistry to prepare simple C-linked disaccharides and C-glycosylated amino acids in order to demonstrate the value of the methodology. We now seek funds to extend the basic methodology to develop a unified route to C-glycosylated targets (such as C-linked oligosaccharides, C-glycosylated peptides and lipids), possibly without the need for protecting groups. The C-glycosylated targets have immense potential in areas as diverse as immunology, cancer chemotherapy, antibacterial chemotherapy and biosensor applications. Known systems will be prepared to illustrate the value of the methodology. We will then design novel compounds in collaboration with Dr. Simon Haseley (CBS, Porton Down) and Prof. Gideon Davies (York). These compounds will be screened as biosensors and enzyme probes, respectively. The effectiveness of these novel systems will be analysed in order to design efficient and selective second generation systems. Please note that this application has MOD support at 50%; the EPSRC request is therefore 85000.
