We propose to develop and use novel spectroscopic techniques to study the structure and vibrational dynamics of proteins, peptides, and nucleic acids. Our main interest is in the determination of the nature of collective vibrational modes in biomolecules, their dynamical properties, their role in biochemical function, and how molecules of biochemical importance (such as inhibitors and intercalators) can modify these properties. Vibrational spectra will be measured using ultrafast optical Kerr-effect (OKE) and infrared spectroscopy, and will be compared with inelastic neutron scattering spectra. We will develop a novel combination of Surface-Enhanced (Resonance) Raman Spectroscopy (SE(R)RS) and OKE that will be used to increase the signal strength. If time allows, we will develop a novel combination of Raman Optical Activity (ROA) and OKE that will be used to identify the chirality of observed modes. We have assembled an international multidisciplinary group of researchers in order to be able to develop new technologies and apply them to an exciting problem in molecular biology.
