The development of new technology for monitoring glucose concentrations in vivo in patients with diabetes mellitus is a worldwide health care priority. Measurement of glucose-induced changes in fluorescent lifetimes (rather than intensity) has several potential advantages over existing methods of glucose sensing and may produce a new generation of stable, accurate and predictable monitors of tissue biochemistry for use in clinical medicine. We intend to prepare fluorophore-labelled glucose-binding proteins, including concanavalin A and glucose oxidase, and test the hypothesis that glucose concentration-dependent changes in near-infrared fluorescent lifetimes of these probes occur which can form the basis of a specific, accurate and precise in vivo glucose assay. Instrumentation will be developed for cuvette and remote, fibre optic-based sensing of time-domain fluorescence using time-correlated single photon counting; the potential interference of light scattering in tissues will be investigated by fluorescence studies in tissue phantoms.
